Non-radioisotopic labels for in situ hybridisation histochemistry: a histochemist's view

چکیده

The early development of in situ hybridisation relied heavily upon the use of radioisotopes and autoradiography for the visualisation of specifically hybridised gene probes (Buongiorno-Nardelli & Amaldi, 1969; Gall & Pardue, 1969, 1971). Radioisotopic labelling retains an important role, especially where maximum sensitivity is required, but in recent years a group of techniques employing non-radioisotopic labelling technology have been developed. They have been designed to avoid the perceived drawbacks of radiolabels in terms of safety and inconvenience. In this review I hope to provide a flavour of the range of non-isotopic techniques available, to discuss their relative merits and to divine future developments. Some general methodologies are outlined but these must be considered as general guidelines from which optimal protocols must be individually determined (some detailed protocols are provided in later chapters).

Detection of abl/bcr Fusion Gene in Patients Affected by Chronic Myeloid Leukaemia by Dual-Colour Interphase Fluorescence in situ Hybridisation

Conventional cytogenetic is the standard technique for detection of Philadelphia (Ph) chromosome in chronic myeloid leukemia (CML). Evaluation of abelson murine leukemia/breakpoint cluster region (abl/bcr) fusion using dual-colour fluorescence in situ hybridization (D-FISH) is an alternative approach allowing rapid and reliable detection of the disease. We employed the technique of interphase D...

Vascular endothelial growth factor mRNA expression in minimal change, membranous, and diabetic nephropathy demonstrated by non-isotopic in situ hybridisation.

AIM To investigate vascular endothelial growth factor (VEGF) mRNA expression in glomerular disease in the context of heavy proteinuria. METHODS Non-radioisotopic in situ hybridisation was performed using a cocktail of 12 deoxyoligonucleotides complementary to VEGF mRNA labelled during solid phase synthesis with 2,4-dinitrophenyl. Archival renal biopsies were studied from cases of minimal chan...

Application of conventional chromosomal aberration and fluorescence in situ hybridisation (FISH) translocation in the assessment of occupationally derived irradiation

Background: Most of our current understanding of the biological effects of exposure to ionising radiation is based on conventional cytogenetic techniques, which enable us to determine the relationship between chromosomal aberration and dose received by radiation workers. However, conventional techniques have numerous limitations and chromosomal aberrations can be easily missed. Since FISH plays...

In situ hybridization and cytochemistry: localization of mRNA at stained neuromuscular junctions with 33P-labeled probes.

We modified the Karnovsky and Roots method of staining sites of acetylcholinesterase (AChE) activity at neuromuscular junctions (NMJs) to survive the lengthy, multiple steps of in situ hybridization and autoradiography. When the original method of Karnovsky and Roots is used to identify the muscle endplates, the stain does not survive the in situ hybridization procedures and association of mRNA...

Non-isotopic in situ hybridisation and immunophenotyping of infected cells in the investigation of human fetal parvovirus infection.

AIMS To compare the use of biotinylated and digoxigenin labelled probes for diagnosis of human fetal parvovirus B19 infection in formalin fixed, paraffin wax embedded tissues; and to assess the cellular distribution of the virus in positive cases. METHODS Sections of lung tissue from 23 cases of anatomically normal non-immune fetal hydrops presenting between 1984 and 1989, and from 13 control...
